Gene Synthesis
- Gene assembly strategies (overlapping, blunt-end, or staggered oligos) have been known for more than two decades.
- Existing barriers - cost and time required for obtaining oligonucleotides; for instance, for a small genome of 5 Mbp, 250,000 oligonucleotides of 40-mers are required. The synthesis
per se would need 60,000 liters of solvents and cost multi-million dollars.
