Characterization of breast cancer using microRNA expression profiling and epigenomic and proteomic measurements in rat model

This month researchers at the Department of Biological Sciences at the University of Lethbridge published the latest in a series of papers describing characterization of breast cancer using microRNA expression profiling and epigenomic and proteomic measurements in rat model.

“Estrogen-Induced Rat Breast Carcinogenesis is Characterized by Alterations in DNA Methylation, Histone Modifications and Aberrant MicroRNA Expression” by Kovalchuk and co-workers

One most important outcome of the current study is the first time demonstration of miRNA expression in mammary tissue correlates with epigenetic dysregulation manifested in morphological changes and alteration of DNA methylation and histone methylation. In particular, using miRNA microarray analysis (provided by LC Sciences) of the E2-induced carcinogenesis development in rat, the down‑regulated miR‑22, miR‑99a, miR‑99b, and miR‑127 are shown to correlate with elevated target oncogene activities.  In this series of studies of the three main components of epigenetics, the authors concluded that “Changes in DNA and histone methylation, and, importantly, deregulation in the miRNAome may lead to the emergence of epigenetically reprogrammed cells with a tumor‑specific phenotype (inhibition of cell cycle arrest and apoptosis, activation of cell proliferation) leading to subsequent malignant transformation”.
